Výzkum metabolismu a metabolického stavu buněk, tkání a organismů, který odpovídá úrovni dnešní vědy, staví na využití "omických" postupů zahrnovaných pod pojem metabolomika.Tato oblast výzkumu vyžaduje vysokou míru odborných znalostí vzhledem k různorodosti struktur metabolitů, komplexitě biologických vzorků a aplikovaných technologií. Regiony Jižní Čechy a Horní Rakousko na jednotlivých pracovištích partnerů disponují významnými odbornými znalostmi v dílčích oblastech metabolického výzkumu, přičemž meziregionální koordinace výzkumných aktivit může vést ke zřetelnému zvýšení efektivity všech zúčastněných partnerů. Proto navrhujeme zřídit dlouhodobou spolupráci mezi Biologickým centrem AV ČR (BC) a Univerzitou Johanna Keplera (JKU) se strategickými regionálními partnery, v jejímž rámci by měla být vytvořena dodatečná laboratorní infrastruktura, která umožní dále rozvíjet výzkum a inovace na špičkové mezinárodní úrovni. Projektová žádost je v souladu s regionálními strategiemi RIS3 a byla přijata Komisí pro inovace Jihočeského kraje do Jihočeského akčního plánu pro inovace, projekt č. 33, Nové metabolomické technologie. Projekt je dále zařazen do akčního plánu "Zdraví – stárnoucí společnost" ve strategickém programu Inovativní Horní Rakousko 2020. Projekt navrhuje zřízení dvou nových komplementárních výzkumných laboratoří, vybavených špičkovými hmotnostními spektrometry pro účely "cílené" a "necílené" metabolomické analýzy. Projekt bude usilovat o dosažení 10 hlavních výstupů: (1) nová meziregionální síť METABO-BL složená pro začátek z 6 regionálních partnerů, (2) efektivnější využití dosavadních znalostí a infrastruktury, (3) zřízení 2 nových komplementárních metabolomických laboratoří v BC a JKU, (4) vytvoření dvou společných výzkumných týmů v laboratořích BC a JKU, (5) společný vývoj nových metabolomických analytických platforem, (6) společné podání výzkumného projektu, (7) ověření konceptu pomocí 3 pilotních výzkumných projektů, (8) zvýšení počtu a úrovně špičkových publikací, (9) Realizace metabolomického kurzu pro studenty, (10) Péče o udržitelný rozvoje sítě METABO-BL. Efektivní využití budované infrastruktury bude zajištěno kooperací s regionálními strategickými partnery: (1) Nemocnice Č. Budějovice a.s., (2) Jihočeská univerzita, (3) Jihočeský vědecko-technický park, (4) Institut lékařské a laboratorní diagnostiky, JKU, (5), Institut bioinformatiky, JKU, (6) Institut pro analytickou chemii, Vídeňská univerzita. Realizace projektu bude zajištěna pomocí 5 pracovních balíčků, nákup vybavení a zprovoznění nových laboratoří (WP I1-I2), trénink obsluhy, implementace stávajících a vývoj nových metod (WP T1-T2 a jejich aplikace v interregionální síti a mezinárodních kooperacích. Description (EN):

A state-of-the-art study of the metabolism and metabolic status of cells, tissues and organisms is building on the application of the "omics" approach to metabolomics. This research field requires a high level of expertise due to the variety of metabolite structures, the complexity of biological samples and the technology used. In the regions of South Bohemia and Upper Austria, individual locations exist which possess expertise in specific sectors of metabolomics research, however, coordination of the activities can lead to a significant increase in efficiency for all stakeholders. Therefore, a key factor seems to be establishing long-term cooperation between the Biology Centre and the Johannes Kepler University, as well as with strategic regional partners, within which a complementary infrastructure can be established that enables international scientific research. The project proposal is in line with the South Bohemian RIS3 strategy and has been implemented in the action plan, project no. 33, “New Metabolomic Technologies”. Furthermore, the project is part of the action field of "Health - Aging Society" in the strategic programme "Innovative Upper Austria 2020". The project involves the construction of two research laboratories equipped with state-of-the-art mass spectrometry for "targeted" and "non-targeted" metabolomics analysis. The following 9 key results are to be achieved in the project: (1) the Interreg network METABO-BL of the 10 partners, (2) efficient use of existing knowledge and infrastructure, (3) the establishment of two new complementary laboratories, (4) the creation of two joint research teams, (5) joint development of new analytical metabolomics platforms, (6) the joint submission of a research project, (7) proof of concept with 3 pilot research projects, (8) an increase of the number of top publications , (9) the metabolomics course for students in the programme area, (10) sustainable development of research cooperation within a European context. A detailed description is provided in Annex C00/I-III, p. 2-5. Efficient use of the established infrastructure will ensure cooperation with regional strategic partners: (1) Budweis Hospital, (2) University of South Bohemia, (3) the South Bohemia Science Technology Park, (4) Institute of Laboratory Diagnostics Kepler Klinikum Linz, (5) Institute for Bioinformatics JKU, (6) Institute for Analytical Chemistry, University of Vienna (see Annex C00/IV, p. 4-8). The implementation of the project is ensured with 5 work packages, the purchase of equipment, the commissioning of the new laboratories (WP I1-I2), the training of the personnel, the implementation of existing and new/in-development methods (WP T1-T2) and their application in the Metabo-BL network as well as international cooperation. The validity of the concept will be tested with 3 pilot projects (WP T3). More in Annex C00.

Eine dem Stand der Wissenschaft entsprechende Erforschung des Stoffwechsels und metabolischen Status von Zellen, Geweben und Organismen baut auf der Anwendung des "omics" Ansatzes Metabolomics auf. Dieses Forschungsgebiet erfordert hohes Expertenwissen auf Grund der Vielfalt der Metabolitstrukturen, der Komplexität von biologischen Proben sowie der angewandten Technologie. In den Regionen Südböhmen-Oberösterreich ist an einzelnen Standorten Expertise in Teilbereichen der Metabolomik-Forschung vorhanden, wobei eine Koordinierung der Aktivitäten zu einer deutlichen Effizienzerhöhung für alle Beteiligten führen kann. Daher erscheint es zielführend, eine langjährige Kooperation zwischen dem Biologiezentrum sowie der Johannes-Kepler-Universität und mit strategischen regionalen Partnern aufzubauen, innerhalb derer eine komplementäre Infrastruktur etabliert werden soll, die international anspruchsvolles wissenschaftliches Arbeiten ermöglicht. Der Projektantrag stimmt mit der Südböhmischen RIS3 Strategie überein und wurde in den Aktionsplan, Projekt No. 33, "Neue metabolomische Technologien" implementiert. Weiters ist das Projekt dem Aktionsfeld "Gesundheit - alternde Gesellschaft" im strategischen Programm Innovatives Oberösterreich 2020 zugeordnet. Das Projekt beinhaltet den Aufbau zweier Forschungslabors ausgestattet mit modernster Massenspektrometrie für "gezielte" und "ungezielte" Metabolomik-Analyse. Im Projekt sollen folgende 9 wichtigste Ergebnisse erzielt werden: (1) das Interreg Netzwerk METABO-BL der 10 Partner, (2) effiziente Nutzung des vorhandenen Wissens und der Infrastruktur, (3) die Einrichtung von zwei neuen komplementären Labors, (4) die Schaffung von zwei gemeinsamen Forschungsteams, (5) die gemeinsame Entwicklung neuer analytischer Metabolomik-Plattformen, (6) die gemeinsame Einreichung eines Forschungsprojekts, (7) Proof-of-Concept mit 3 Pilotforschungsprojekten, (8) eine Erhöhung der Anzahl der Top-Publikationen, (9) Der Metabolomik-Kurs für Studenten im Programgebiet, (10) nachhaltige Entwicklung der Forschungszusammenarbeit im europäischen Kontext. Eine detaillierte Darstellung ist in Anhang C00/I-III, S.2-5 gegeben. Die effiziente Nutzung der aufgebauten Infrastruktur wird die Zusammenarbeit mit regionalen strategischen Partnern gewährleisten: (1) Krankenhaus Budweis, (2) Südböhmische Universität, (3) der Wissenschafts-Technologiepark Südböhmen, (4) Institut für Labordiagnostik Kepler Klinikum Linz, (5) Institut für Bioinformatik JKU, (6) Institut für Analytische Chemie, Universität Wien (siehe Anhang C00 /IV, S.4-8). Die Umsetzung des Projekts wird durch 5 Arbeitspakete sichergestellt, den Kauf von Ausrüstung, die Inbetriebnahme der neuen Laboratorien (WP I1-I2), die Ausbildung des Personals, die Umsetzung bestehender und Entwicklung neuer Methoden (WP T1-T2) und ihre Anwendung im Metabo-BL Netzwerk sowie internationale Zusammenarbeit. ● Schaffung eines interregionalen Wissen-Netzwerkes Metabo-BL mit mindestens 6 regionalen strategischen Partnern (Personal, Labor, Instrumentierung, Know how, excellente Forschung und Innovation) ● Einrichtung zweier komplementärer Metabolomiklabors: Labor 1 für hochempfindliche gezielte Analytik im Budweis (BC Partner) um Unterschiede zwischen Kontrollgruppen (zum Beispiel gesunde Patienten) und Zielgruppen (Krankheitsstadium ) zu untersuchen Labor 2 für ungezielte Analytik wie das Screening und die Identifizierung von Metaboliten in den Kontrol- und Zielgruppen ● Schaffung eines gemeinsamen Forschungsteams mit neuen Stellen für junge talentierte Forscher ● Proof-of-Concept aufgrund der 3 Pilotforschungsprojekten und Erhöhung der Zahl an gemessenen Metaboliten ● Anwendung der neuen Metabolomikplattformen in Forschung mit regionalen Partnern und im europäischen Kontext (medizinische Chemie, personalisierte Medizin, klinische Diagnostik usw) Anhang C00/II-III.
