MATEGRA: Advanced porous biomaterials functionalised with stem cells for improved osteointegration of implants

The MATEGRA project focuses on the development, optimisation and testing of new types of biocompatible surfaces of titanium implants for improved osteointegration and bone tissue adhesion. The intention is to explore the know-how in the production of new biomaterials and porous surfaces for the production of model samples for biological compatibility. The main objectives of the project are the cooperation of research institutions and professional groups, as well as the transfer of knowledge in the development and testing of new porous biomaterials. The Research Center — New Technology (NTC) as part of the University of West Bohemia (UWB) and trauma surgery of the University Hospital Regensburg (UKR) form a joint scientific research platform for the development of new porous biomaterials and biological activity testing. The research objectives of the project can be divided into several steps: A) the production of a coarse poring titanium surface, which has been structurally modified by laser irradiation B) Incorporation of sol-gel polymerised titanium and silicon precursors into these surfaces to create coarse porous two-phase titanium silicate structures Laser-induced insertion of selected bioactive nanoparticles (SiOx, TiOx, and their hydrated forms) to stimulate bone growth D) Incorporation of hyroxyapatite particles into these structures, which are intended to serve as origins for bone growth Another goal of the project is to test the biocompatibility of the manufactured materials and to check the suitability of the new materials for clinically relevant in vivo bone repair models. The research objectives of this section are: The cultivation of primary and genetically marked (GFP, green fluorescent protein) mesenchymal cells, which are precursor cells of the skeleton and primary osteoblasts (differentiated bone cells) B) Testing the biocompatibility of the material through cell distribution and accumulation analyses C) Testing of cell survival and cell propagation on these materials D) Investigation of the cell response in relation to stem cell differentiation towards osteoblasts, gene and protein expression The project requires cooperation between the two institutions, as the problem in the fields has not yet been solved. The added value of the project lies in problem-solving through close cooperation between the research teams and their know-how on the development of new functional biomaterials, as well as the promotion of scientific activities in the field of materials engineering and regenerative medicine.

Projekt MATEGRA je zaměřen na vývoj, optimalizaci a testování nových typů biokompatibilních povrchů u titanových implantátů pro zlepšení osseointegrace a adheze kostní tkáně. Záměrem je získat know-how pro přípravu nových porézních biomateriálů a realizovat výrobu modelových vzorků pro testování biologické kompatibility. Hlavními cíli projektu jsou propojení výzkumných institucí, odborných skupin a transfer znalostí v problematice vývoje a testování nových porézních biomateriálů. Výzkummé Centrum - Nové Technologie (NTC) jako součást Západočeské Univerzity v Plzni (ZČU) a Univerzitní Klinika a Úrazová Poliklinika v Regensburgu (UKR) vytvoří společnou vědecko-výzkumnou platformu se zaměřením na přípravu nových porézních biomateriálů a jejich testování biologické aktivity. Výzkumné cíle projektu lze rozdělit do několika kroků: A) Výroba makroporézních povrchových vrstev titanu a jejich strukturování pomocí indukovaného laserového záření; B) Inkorporace rozpustných prekurzorů titanu a oxidu křemičitého (metodou sol-gel) do těchto vrstev za účelem dosažení makroporézních bikontinuálních titanosilikátových struktur; C) Laserové inkorporace vybraných bioaktivních nanočástic (SiOx, TiOx, jeho hydratovaných forem) do těchto struktur katalyzujících růst kostní tkáně; D) Začlenění hydroxyapatitových zrn do těchto struktur, které budou sloužit jako zárodečná místa pro růst biologické tkáně.   Další cíl projektu je zhodnocení biokompatibility připravených materiálů a ověření vhodnosti nových materiálů pro klinicky relevantní in vivo modely opravy kostí. Výzkumné cíle této části jsou: A) Kultivace primárních a geneticky značených mezenchymálních kmenových buněk GFP (zelený fluorescenční protein), které jsou prekurzorovými buňkami kostry a primárními osteoblasty; B) Zkoumání biokompatibility materiálů prostřednictvím analýz buněk a studium jejich adheze k vytvořeným substrátům; C) Identifikace přežití a propagace buněk na materiálech; D) Zkoumání buněčné odezvy na materiály a studium exprese genové a proteinové diferenciace.   V rámci projektu bude probíhat spolupráce obou institucí na tématu, které nebylo v regionech doposud řešeno. Přidaná hodnota projektu je očekávána v rozvoji nové problematiky, úzkém propojení výzkumných týmů, získání potřebného know-how k přípravě nových funkčních biomateriálů a v popularizaci vědecké činnosti v oblasti materiálového inženýrství a regenerativní medicíny.

Das MATEGRA Projekt konzentriert sich auf die Entwicklung, Optimierung und Erprobung neuer Arten von biokompatiblen Oberflächen von Titanimplantaten zur verbesserten Osteointegration und Knochengewebe-Haftung. Die Absicht ist es, das Know-how für die Herstellung von neuen Biomaterialien und porösen Oberflächen zur Herstellung von Modellproben für biologische Verträglichkeit zu erforschen. Die Hauptziele des Projektes sind die Zusammenarbeit der Forschungseinrichtungen und Berufsgruppen, sowie der Transfer von Wissen in der Entwicklung und Erprobung neuer poröser Biomaterialien. Hierbei bilden das Research Center - New Technology (NTC) als Teil der University of West Bohemia (UWB) und die Unfallchirurgie des Universitätsklinikums Regensburg (UKR) eine gemeinsame wissenschaftliche Forschungsplattform für die Entwicklung der neuen porösen Biomaterialien und der biologischen Aktivitätstestung. Die Forschungsziele des Projekts lassen sich in mehrere Schritte unterteilen: A) die Produktion einer grobporingen Titan-Oberfläche, welche mittels Laserbestrahlung strukturell modifiziert wurde B) Einbringen von sol-gel polymerisierten Titan- und Silicium-Vorläufern in diese Oberflächen, um grobporige zweiphasige Titansilikat-Strukturen zu schaffen C) laser-induziertes Einbringen von ausgewählten bioaktiven Nanopartikeln (SiOx, TiOx, und deren hydratisierten Formen), welche das Knochenwachstum anregen sollen D) Einbringen von Hyroxyapatit-Teilchen in diese Strukturen, welche als Ursprung für das Knochenwachstum dienen sollen Ein weiteres Ziel des Projekts ist die Testung der Biokompatibilität der hergestellten Materialien und die Überprüfung der Eignung der neuen Materialien für klinisch relevante in-vivo-Modelle zur Knochenreparatur. Die Forschungsziele dieses Abschnitts sind: A) die Kultivierung von primären und genetisch markierten (GFP, green fluorescent protein) Mesenchymalzellen, welche Vorläuferzellen des Skeletts und von primären Osteoblasten (differenzierte Knochenzellen) sind B) Testung der Biokompatibilität des Materials durch Zellverteilungs- und Anlagerungs-Analysen C) Testung des Zell-Überlebens und der Zellausbreitung auf diesen Materialien D) Untersuchung der Zellantwort in Bezug auf die Stammzell-Differenzierung in Richtung Osteoblasten, Gen- und Protein-Expression Das Projekt benötigt die Zusammenarbeit zwischen den beiden Institutionen, da die Problematik auf den Gebieten bisher nicht gelöst ist. Der Mehrwert des Projektes liegt bei der Problembehandlung durch enge Zusammenarbeit zwischen den Forschungsteams mit deren Know-how zur Entwicklung neuer funktioneller Biomaterialien, sowie der Bekanntmachung von wissenschaftlichen Aktivitäten auf dem Gebiet der Werkstofftechnik und regenerativer Medizin.

Stěžejním výsledkem projektu bude vytvoření hierarchicky porézního biomateriálu se zvýšenou osseointegrací. Daného cíle bude dosaženo laserovým vytvořením porézních Ti povrchů, které budou dále vyplňovány sol-gel metodou bioaktivními skly a tento komposit bude obohacen bioaktivními nanočásticemi deponovanými pomocí pulsní laserové depozice. Na takto vzniklém novém typu implantátu budou prováděny in vitro testy růstu buněk. Zapojením dvou týmu do dané problematiky dojde k posílení vazeb mezi regiony a k vytvoření unikátního týmu v dané problematice, který v rámci česko-bavorského regionu zatím chybí. Mezi kvantifikovatelnými výstupy lze jmenovat celkově 4 workshopy (2 studentské a 2 odborné) a alespoň jednu společnou publikaci v mezinárodním vědeckém časopise. Další výsledek projektu představuje také vhodně zvolená duševní ochrana získaných znalostí, která bude rovným dílem rozdělena mezi obě spolupracující instituce. Optimalizovaný materiál, znalostní báze jeho přípravy a ověření možnosti aplikace formou implantátu jsou očekávané výsledky projektu, na kterých lez postavit pokračující spolupráci mezi regiony.

Ein wichtiges Ergebnis des Projektes ist die Herstellung eines porösen Biomaterials mit einer verbesserten Osteointegration. Durch Laserbehandlung soll eine poröse Ti-Oberfläche erreicht werden, die durch Sol-Gel bioaktive Gläser und den Verbund von bioaktiven Nanopartikel über gepulste Laserabscheidung verbessert wird. Mit dieser Technologie wird eine neue Art von Implantaten geschaffen und mit Hilfe verschieden in-vitro-Zelltests wird die Biokompabilität überprüft. Durch die zwei Forschungsteams wird die Beziehung zwischen den Regionen gestärkt und eine Zusammenarbeit geschaffen, die bisher in der tschechisch-bayerischen Region einmalig ist. Es können insgesamt vier Workshops (zwei für Studenten und zwei für fertig ausgebildete Wissenschaftler) und mindestens eine gemeinsamen Veröffentlichung in einer wissenschaftlichen Zeitschrift aus diesem Projekt entstehen. Ein weiteres verfolgtes Ziel ist die Patentierung der aus dem Projekt resultierenden Ergebnisse, welches zu gleichen Teilen zwischen den beiden Kooperations-Institutionen aufgeteilt wird. Die zu erwartenden Ergebnisse aus diesem Projekt (optimiertes Material, Verifizierung der Einsatzmöglichkeiten in Form des neuen Implantates) sind die Basis auf denen neue Kooperationen zwischen den Regionen aufgebaut werden können.
