La microbiota intestinal es un elemento importante y poco reconocido en la disfunción neurocognitiva de sujetos de edad avanzada. La prevalencia de trastornos neurodegenerativos se encuentra entre las mayores amenazas actuales para el envejecimiento en EU. Fuerte evidencia destaca los efectos de la obesidad en la disfunción neurocognitiva y en la composición de la microbiota. Sin embargo, el conocimiento es aún muy escaso en humanos. Para abordar estos problemas, se establecerá un consorcio de expertos transfronterizo en el área, con herramientas exploratorias y bases de datos humanas, aprovechando el estudio de cambios paralelos en la composición corporal, microbiota intestinal, metabolómica sérica, microestructura cerebral y cognición. Se examinará la transcriptómica tisular, la metabolómica del suero y la orina y la metagenómica en asociación con la cognición, transversalmente y longitudinalmente en cohortes con más de 1.700 sujetos. Usando modelos bioinformáticos y biomatemáticos, se descubrirán especies bacterianas y sus metabolitos que interactúan con el eje microbiota intestinal-colon-hígado-cerebro, y su impacto en el declive neurocognitivo. El conocimiento sobre el modo de acción se obtendrá en ratones libres de gérmenes colonizados con comunidades bacterianas en las que se estudiarán procedimientos neurocognitivos extensos después del trasplante fecal. La información se utilizará para construir herramientas para predecir individualmente y prevenir enfermedades asociadas con microbiota intestinal-colon-hígado-cerebro.

Gut microbiota is an important and under-recognized player in neurocognitive dysfunction of aged subjects. The prevalence of neurodegenerative disorders are among the greatest current health threats of aging in EU. Strong evidence highlights the effects of obesity on neurocognitive dysfunction and altered microbiota composition. However, the knowledge is yet very scarce in humans. To overcome these issues, we will set a unique consortium of cross-border experts in the field, with exploratory tools, and human databases, taking advantage of the study of parallel changes in body composition, gut microbiota, serum metabolomics, brain microstructure and cognition. We will also scrutinize tissue transcriptomics, serum and urine metabolomics, and metagenomics in association with cognition, both cross-sectionally and longitudinally in cohorts involving >1700 subjects. Bioinformatics and biomathematics models will uncover bacterial species and their metabolites interacting with the gut microbiota-colon-liver-brain axis, and its impact on neurocognitive decline. Gain of knowledge regarding mode of action will be obtained in germ-free mice colonized with ad hoc bacterial communities in which extensive neurocognitive procedures will be studied after fecal transplantation. The information will also be used to build tools that will permit to individually predict and prevent diseases associated with gut microbiota-colon-liver- brain.

Le microbiote intestinal est un élément important et peu reconnu du dysfonctionnement neurocognitif des personnes âgées. La prévalence des maladies neurodégénératives est l’une des plus grandes menaces au vieillissement en EU. Des preuves solides mettent en évidence les effets de l'obésité sur le dysfonctionnement neurocognitif et sur la composition du microbiote. Cependant, la connaissance est encore très rare chez l'homme. Pour résoudre ces problèmes, un consortium d'experts transfrontaliers sera créé dans la région, avec des outils d'exploration et des bases de données humaines, tirant parti de l'étude des modifications parallèles de composition corporelle, microbiote intestinal, métabolomique sérique, microstructure cérébrale et cognition. La transcriptomique tissulaire, métabolomique du sérum et urine et métagénomique en association avec cognition, transversalement et longitudinalement dans les cohortes de >1 700 sujets, seront examinées. À l'aide de modèles bioinformatiques et biomathématiques, nous découvrirons des espèces bactériennes et leurs métabolites qui interagissent avec l'axe microbiota-colon-foie-cerveau intestinal, ainsi que leur impact sur le métabolisme glycémique et le déclin neurocognitif. Des connaissances sur le mode d'action seront obtenues chez des souris exemptes de germes colonisées par des communautés bactériennes dans lesquelles de nombreuses procédures neurocognitives seront étudiées après une transplantation fécale. L'information sera utiliséepour créer des outils permettant de prévoir individuellement et de prévenir les maladies associées au microbiote intestinal-colon-foie-cerveau.

Concluye el proyecto Thinkgut alcanzando con éxito todos sus objetivos: ThinkGut llega a su fin consolidándose como un proyecto transfronterizo clave en el desarrollo del estudio del papel de la microbiota intestinal en los trastornos neurocognitivos asociados a la edad avanzada y a la obesidad. La prevalencia de trastornos neurodegenerativos es una de las amenazas actuales más importantes para el envejecimiento en Europa. Se sabe que la obesidad tiene efectos sobre la neurocognición y composición de la microbiota intestinal, siendo, además, la microbiota un elemento tan importante como poco reconocido en la disfunción neurocognitiva en la edad avanzada. Dos años después de su inicio, y llegada la fecha de cierre del proyecto, ThinkGut ha podido llevar a cabo y desarrollar todos los objetivos y actividades que se habían marcado en su inicio relacionados con este ámbito de investigación. En este sentido, el ThinkGut ha logrado con éxito grandes avances como herramientas diagnóstico-terapeúticas basadas en el microbioma o la creación de un repositorio anonimizado transfronterizo con información clínico-biológica sobre la relación microbiota-cognición para el desarrollo de modelos de biología de sistemas multi-escala para el eje intestino-cerebro en ratones humanizados y humanos. Gracias al proyecto, también se han podido formar perfiles de investigación especializados en la interrelación que existe entre la microbiota y la cognición dentro del territorio transfronterizo. Los resultados finales de este proyecto son una colaboración sinergética y estratégica con centros de investigación del territorio transfronterizo que permitirán la creación de herramientas para la prevención, el diagnóstico y el tratamiento de los trastornos neurocognitivos asociados en la edad avanzada y la obesidad, respondiendo así a una de las problemáticas más destacadas sobre envejecimiento en la Unión Europea.

Achievements (EN):

It concludes the Thinkgut project successfully achieving all its objectives: ThinkGut comes to an end by consolidating itself as a key transboundary project in the development of the study of the role of the gut microbiota in neurocognitive disorders associated with advanced age and obesity. The prevalence of neurodegenerative disorders is one of the most important current threats to ageing in Europe. It is known that obesity has effects on neurocognition and composition of the intestinal microbiota, being, in addition, the microbiota an important element as well as little recognised in neurocognitive dysfunction in advanced age. Two years after its inception, and the closing date of the project arrives, ThinkGut has been able to carry out and develop all the objectives and activities that had been marked in its beginning related to this field of research. In this sense, the ThinkGut has successfully achieved great advances as diagnostic-therapeutic tools based on the microbiome or the creation of a transboundary anonymised repository with clinical-biological information on the microbiota-cognition relationship for the development of multi-scale biology models for the gut-brain axis in humanised and human mice. Thanks to the project, it has also been possible to form research profiles specialised in the interrelationship between microbiota and cognition within the cross-border territory. The final results of this project are a synergetic and strategic collaboration with research centers in the cross-border territory that will allow the creation of tools for the prevention, diagnosis and treatment of associated neurocognitive disorders in the elderly and obesity, thus responding to one of the most prominent problems on aging in the European Union.
