Description (EN):

In this project, an eco-innovative process will be developed that addresses the reuse of wastewater for resource-efficient waste management and an alternative process for the production of bioplastics, for increased availability and concomitant contribution to environmental protection. Nutrients are used from various wastewaters from dairies or municipal wastewater treatment plants to produce polyhydroxybutyrate (PHB). PHB is a biodegradable polymer that is naturally produced by microorganisms. These include cyanobacteria (microalgae), which sustainably produce PHB through photosynthesis using light and CO2. The nutrients required by the bacteria for the growth and formation of PHB are to be provided cost-effectively in part by adding wastewater from dairies or sewage treatment plants. The production of PHB is currently only realized by a few companies on a large scale and mostly on a petroleum basis, making the market price very high and active use still too expensive for packaging industries. For this reason, alternative production processes for PHB are urgently needed. Therefore, the aim of this project is to establish an innovative process for the cultivation of cyanobacteria in wastewater-containing medium for the production of PHB on a pilot scale. The PHB production process will be available to companies and interested parties to increase the use of bioplastics. The research results will be published in peer-reviewed journals. The project is realized through the cooperation of 3 project partners, each with unique knowledge in the border region. The Czech lead partner MBU is an internationally recognized center for basic and applied research on microalgae and cyanobacteria, including algal biotechnology. Therefore, MBU will be responsible for the cultivation process, addition and, if necessary, treatment of wastewater, and optimization of PHB production on a pilot scale. The expertise of the FH OOE in the field of genetic engineering of phototrophic microorganisms will be used to increase the PHB production of the cyanobacteria and to improve the utilization of certain nutrients from the wastewater. The extensive knowledge of TU Vienna regarding laboratory scale cultivation of microorganisms and analysis of various substances from the biomass, will be used in this project to screen the transformants produced by FH OOE for the amounts of PHB produced and to develop a method to isolate PHB from the biomass. The selected transformants will then be cultivated at MBU on a pilot scale for PHB production, the biomass will be processed, and the PHB will be extracted at TU Wien. This produced and structurally unchanged PHB can then be used for various applications, e.g. for bioplastic production.

In diesem Projekt soll ein öko-innovativer Prozess entwickelt werden, welcher die Weiterverwendung von Abwässern für eine ressourceneffiziente Abfallwirtschaft und ein alternatives Verfahren zur Produktion von Bioplastik, für eine erhöhte Verfügbarkeit und einhergehenden Beitrag zum Umweltschutz, adressiert. Nährstoffe werden aus verschiedenen Abwässern von Molkereien oder kommunalen Kläranlagen zur Herstellung von Polyhydroxybutyrat (PHB) verwendet. PHB ist ein biologisch abbaubares Polymer, dass von Mikroorganismen natürlicherweise gebildet wird. Dazu gehören Cyanobakterien (Mikroalgen), welche durch Photosynthese PHB unter Verwendung von Licht und CO2 nachhaltig produzieren. Die von den Bakterien für das Wachstum und die Bildung von PHB benötigten Nährstoffe sollen teilweise durch Zugabe von Abwässern der Molkereien oder Kläranlagen kostengünstig bereitgestellt werden. Die Produktion von PHB wird derzeit nur von wenigen Firmen großtechnisch und meist auf Erdölbasis realisiert, wodurch der Marktpreis sehr hoch und eine aktive Nutzung für Verpackungsindustrien noch zu teuer ist. Aus diesem Grund sind alternative Produktionsprozesse für PHB dringend erforderlich. Daher ist das Ziel dieses Projekts ein innovatives Verfahren zur Kultivierung von Cyanobakterien in abwasserhaltigem Medium für die Produktion von PHB im Pilotmaßstab zu etablieren. Der PHB-Produktionsprozess soll Firmen und Interessenten zur Verfügung stehen, um die Nutzung von Bioplastik zu erhöhen. Die Forschungsergebnisse werden in Fachjournalen veröffentlicht. Das Projekt wird durch die Kooperation von 3 Projektpartnern realisiert, die jeweils über einzigartiges Wissen in der Grenzregion verfügen. Der tschechische Leadpartner MBU ist ein international anerkanntes Zentrum für Grundlagen- und angewandte Forschung an Mikroalgen und Cyanobakterien, einschließlich der Algenbiotechnologie. Daher wird das MBU für den Kultivierungsprozess, die Zugabe und gegebenenfalls Aufbereitung der Abwässer und die Optimierung der PHB-Produktion im Pilotmaßstab zuständig sein. Die Expertise der FH OOE im Bereich der Gentechnik phototropher Mikroorganismen wird genutzt, um die PHB Produktion der Cyanobakterien zu steigern und die Verwertung bestimmter Nährstoffe aus den Abwässern zu verbessern. Das umfangreiche Wissen der TU Wien hinsichtlich der Kultivierung von Mikroorganismen im Labormaßstab und Analyse verschiedener Substanzen aus der Biomasse, wird in diesem Projekt zum Screening der von der FH OOE hergestellten Transformanten hinsichtlich der produzierten PHB Mengen und zur Entwicklung einer Methode zur Isolierung von PHB aus der Biomasse genutzt. Die ausgewählten Transformanten werden dann am MBU im Pilotmaßstab zur PHB Herstellung kultiviert, die Biomasse aufgearbeitet und das PHB an der TU Wien extrahiert. Dieses produzierte und strukturell unveränderte PHB kann dann für verschiedene Applikationen zB. zur Bioplastikproduktion verwendet werden.

V tomto projektu bude vyvinut inovativní ekologicky proces, který se bude zabývat opětovným zpracováním odpadních vod, a tedy využíváním odpadních zdrojů a novým postupem výroby bioplastů, což povede k jejich dostupnosti a zároveň přispěje k ochraně životního prostředí. K výrobě polyhydroxybutyrátu (PHB) budou použity živiny pocházející z různých druhů odpadních vod mlékárenské výroby nebo z komunálních čistíren odpadních vod. PHB je biologicky odbouratelný polymer, který je přirozeně produkován mikroorganismy. Patří mezi ně sinice (fotosyntetizující bakterie, mikrořasy), které jsou schopny prostřednictvím fotosyntézy, za využití světla a CO2, udržitelně produkovat PHB. Živiny potřebné pro růst a tvorbu PHB budou sinice získávat z odpadních živin z mlékáren nebo komunálních čistíren odpadních vod. Výroba PHB v průmyslovém měřítku je v současné době prováděna pouze několika společnostmi, především pro využití při výrobě obalových materiálů, většinou je založena na ropné bázi, tedy neudržitelná a nákladná. Alternativní a udržitelné výrobní procesy PHB proto považujeme za žádoucí. Cílem tohoto projektu je představit inovativní proces výroby PHB v pilotním měřítku pomocí pěstování sinic v médiu obsahujícím odpadní vodu. Proces výroby PHB bude k dispozici společnostem a zúčastněným stranám, což může přispět ke zvýšenému používání bioplastů a rozvoji bioekonomiky. Výsledky výzkumu budou rovněž publikovány v odborných časopisech. Projekt bude realizován třemi projektovými partnery v přeshraničním regionu, z nichž každý má jedinečné znalosti. Hlavní český partner, Mikrobiologický ústav AV ČR (MBU), je mezinárodně uznávané centrum základního a aplikovaného výzkumu mikrořas a sinic, včetně biotechnologií řas. Proto bude MBU odpovědný za proces pěstování biomasy s využitím odpadní vody a za optimalizaci produkce PHB v pilotním měřítku. Odbornost Fachhochschule Oberösterreich (FH OOE) v oblasti genetického inženýrství fototrofních mikroorganismů bude využita ke zvýšení produkce PHB u sinic a k lepšímu využití určitých živin z odpadních vod. Rozsáhlé znalosti Technické univerzity ve Vídni (TU Wien), které se týkají kultivace mikroorganismů v laboratorním měřítku a analýzy různých látek v biomase, budou v tomto projektu použity ke screeningu geneticky vylepšených sinic vytvořených pracovištěm FH OOE s ohledem na vyprodukované množství PHB a k vývoji extrakční metody pro izolaci PHB z biomasy. Vybrané kmeny produkujíci PHB budou kultivovány na pracovišti MBU v pilotním měřítku a PHB ze zpracované biomasy bude extrahováno na pracovišti TU Wien. Takto získaný PHB, bude možné použít v různých aplikacích, například pro výrobu bioplastů.

Achievements (EN):

The project has succeeded in developing a cyanobacterial cultivation process where the substrate is wastewater. Two types of wastewater were used for the growth of cyanobacterial biomass (i) municipal wastewater from the treatment process and (ii) wastewater from dairy production. Both substrates are sources of nitrogen (250 mg/L for both substrates) and phosphorus (150 mg/L municipal wastewater and 25 mg/L dairy wastewater), the main components required for cyanobacterial growth. A strain of the cyanobacterium Synechocystis generated by UV mutagenesis with higher PHB production was used for cultivation. A thin-layer outdoor cultivation unit was used for the cultivation in the wastewater of the municipal sewage treatment plant. Cultivation in dairy wastewater was carried out in a closed annular photobioreactor. This was performed to optimize conditions for later testing a genetically engineered strain with higher PHB production and lactose processing ability. Cultivating the strain in both pilot-scale units took 4-6 weeks to achieve the maximum amount of PHB. On the day of biomass harvest, PHB content was 23 % of the dry cell weight in municipal wastewater and 10 % in dairy wastewater. Compared with available data, PHB production in the phototrophic regime (no organic carbon source) of genetically unmodified cyanobacteria is 4-25% under optimal culture medium conditions in pilot-scale units. The strain used in the Plastocyan project achieved 37% PHB cell dry weight under laboratory conditions on a small scale in the optimal growth substrate in the previous study. However, the main benefit of the project is the recycling of wastewater. The results of the project are also above average in this respect. In both types of wastewater, it was possible to grow biomass up to a density of 2-2.5 g/L, while the average values of cyanobacterial growth in wastewater are reported to be 0.6-3.15 g/L. PHB granules can be extracted from the cells by various extraction methods, mainly using organic solvents such as chloroform, which can be hazardous to the environment. The project also addresses the environmentally and economically friendly alternative of extracting PHB from biomass using so-called ionic liquids belonging to the category of green solvents. In many cases, extracting the product using ionic liquids is more accessible, can be recycled, and therefore reused. The project is also developing a genetically modified strain that can overproduce PHB and use lactose as a carbon source. This could streamline PHB production in closed cultivation units using dairy wastewater. The UV-generated mutant Synechocystis sp. PCC 6714 Mt_a24 was genetically improved by eliminating two genes (spsA and glgC) related to competing metabolic pathways. This elimination is supposed to trigger a metabolic shift towards an increased PHB production rate. At the same time, genes enabling lactose utilization (beta-Gal) as a carbon source were cloned and successfully introduced in the cyanobacteria. Moreover, work focusing on increasing PHB production (cloning of plasmids containing phaA and phaB genes) is still ongoing. The main results achieved in the project include: a) Optimization of cyanobacteria cultivation in wastewater (urban wastewater, dairy wastewater) for the production of bioplastic - polyhydroxybutyrate (PHB) on a pilot scale. b) Ecological extraction of PHB from cyanobacterial biomass using ionic liquids. c) Generating genetically improved strains with the potential for higher PHB production and utilization of lactose for their growth.

Das Hauptergebnis des Projekts wird der Kultivierungsprozess von Cyanobakterien unter Verwendung von Abwässern als Nährstoffquellen für die biotechnologische Produktion von PHB sein. Die Verwendung von Abwässern aus verschiedenen Quellen zur Herstellung geeigneter Kultivierungsmedien für das Wachstum von Cyanobakterien und der Aufbau der Kultivierungstechnologie für die PHB-Produktion im Pilotmaßstab werden ein Teil der Projektergebnisse sein. Ein weiterer Output stellen genetisch optimierte Cyanobakterienstämme dar, die eine erhöhte Produktion von PHB aufweisen. Darüber hinaus sollen Cyanobakterienstämme selektiert werden, die in Molkereiabwässern enthaltene Laktose abbauen und verwerten, um diese Reststoffe für die PHB-Produktion zu nutzen. Abschließendes Ergebnis wird die Aufreinigung des produzierten PHB aus den Cyanobakterienzellen sein, um einen vollständigen Prozess von der Produktbildung bis zu einer späteren Verwendung zu erreichen.

Hlavním výsledkem projektu je kultivační proces sinic využívající odpadní vodu jako zdroj živin pro biotechnologickou výrobu PHB. Součástí výstupů projektu bude technologie zpracování odpadní vody z různých zdrojů pro výrobu vhodných kultivačních médií a zavedení kultivační technologie pro produkci PHB v pilotním měřítku. Dalším výstupem jsou geneticky optimalizované kmeny sinic, které vykazují zvýšenou produkci PHB. Kromě toho budou vybrány kmeny sinic, které degradují a využívají laktózu obsaženou v mlékárenské odpadní vodě, aby ji využily pro výrobu PHB. Konečným výsledkem bude PHB získaná extrakcí z buněk sinic za účelem dosažení úplného procesu od tvorby produktu po jeho následné použití.
