The objective of IMPROVE-STEM is to develop an integrated set of tools necessary for the amplification of mesenchymal stem cells to promote their application in cell therapy in our hospitals. These tools will be based on the adoption of microbeads whose surface will be optimised to ensure control of cell adhesion and the design of a bioreactor whose operating conditions will be adapted for the cultivation of adhering stem cells on microbeads. Monitoring of cell behaviour will be carried out to ensure the quality, homogeneity and purity of the cells. The involved multidisciplinary consortium brings together advanced skills in materials science, bioprocess engineering and cell biology. At the level of the Greater Region, it forms the basis of a Platform of Excellence in the culture of mesenchymal stem cells.

L’objectif d’IMPROVE-STEM vise au développement d’un ensemble intégré d’outils nécessaires à l’amplification de cellules souches mésenchymateuses pour promouvoir leur application en thérapie cellulaire dans nos hôpitaux. Ces outils reposeront sur l’adoption de microbilles dont la surface sera optimisée pour assurer un contrôle de l’adhésion des cellules et sur le design d’un bioréacteur dont les conditions opératoires seront adaptées pour la culture de cellules souches adhérentes sur microbilles. Un suivi du comportement des cellules sera réalisé pour en garantir la qualité, homogénéité et pureté des cellules. Le consortium multidisciplinaire impliqué rassemble des compétences pointues en sciences des matériaux, en génie des bioprocédés et en biologie cellulaire. A l’échelle de la Grande Région, il constitue le socle d’une Plateforme d’Excellence dans la culture de cellules souches mésenchymateuses.

Das Forschungsprojekt brachte wesentliche Fortschritte auf dem Gebiet der Expansionsprozesse mesenchymaler Stammzellen und deren Aufskalierung. Es wurden drei innovative, sich ergänzende Werkzeuge entwickelt. Dies sind: 1. Mikrobeads mit einer kontrollierten Oberfläche, die das Wachstum von Stammzellen fördern und ihre Gewinnung nach der Expansion erleichtern, 2. ein Rührkessel-Bioreaktor, dessen Design und Betriebsbedingungen für die Expansion von an Mikrobeads haftenden Stammzellen optimiert sind, und 3. ein Charakterisierungs- und Validierungsprotokoll für die Qualität und Eigenschaften der produzierten Stammzellen.

Achievements (EN):

The research project brought significant progress in the field of the expansion processes of mesenchymal stem cells and their scaling. Three innovative, complementary tools have been developed. These are: 1. Microbeads with a controlled surface that promote the growth of stem cells and facilitate their recovery after expansion; 2. a stirring boiler bioreactor whose design and operating conditions are optimised for the expansion of stem cells adhering to microbeads, and 3. a characterisation and validation protocol for the quality and properties of the stem cells produced.
