Analysis and correlation between epigenetics and brain activity to assess the risk of chronic and episodic migraine in women

Objectives: The general objective of the project is to design a tool based on the analysis of epigenetics (by means of blood samples) and brain activity (through electroencephalogram, EEG), which characterises chronic and episodic migraine, simplifying and improving its treatment. This tool will be built from the analysis of blood samples and records of the electrical activity of the brain. Both are rapid and non-invasive or minimally invasive sampling techniques for the patient. In the scientific literature, several research focused on the genetics of migraine patients can be found. However, epigenetics has hardly been studied, that is, the mechanisms of regulation of gene expression without modification of the genetic code. Such regulatory mechanisms are highly influenced by environmental factors, which could also be related to changes in neuronal activity. Therefore, it would be very novel to study the changes that migraine produces in the electrical activity of the brain, which is recorded through the EEG and to relate these changes to the epigenetics. The tool developed will base its accuracy on the correlation and joint information between epigenetic information and the electrical activity of the brain. This research aims to facilitate labour integration and equal social inclusion. In addition, networking and cooperative relationships between cross-border entities will be encouraged to identify the underlying epigenetic and neural substrates of migraine. The participating entities will benefit from the possible funding to improve their current equipment aimed at dealing with migraine patients, as well as the processing and analysis of samples. On the other hand, they will benefit from hiring human resources and specialised training of them in the study of this disease. In relation to the specific objective set out by the programme, it is expected that the creation of the partnership will lay the foundations for a network of scientific excellence in the study of migraine. In this regard, the dissemination of the results derived from the project in national and international congresses, as well as the publication of articles in indexed international journals, will improve the prestige and competitiveness of the participating entities individually and as a partnership. In addition, this project also considers opening additional avenues of cooperation between participating entities for future studies, such as those related to the treatment of this widespread disease in our regions. Results: The overall results of the project are as follows: — Increase in the number of researchers in the region of Norte de Portugal and Castilla y León. It is intended to hire at least four new researchers thanks to the funding provided by this call. The new staff will be fully dedicated to the project, encouraging interaction between the participating entities, which will be carried out through visits and stays in them. — Improvement of the equipment necessary for the study of migraine in the participating entities: computer equipment, portable electroencephalograph and laboratory equipment (first, methylation analysis kits, PCR kits, qPCR and RT-PCR, etc.). — Creation of a new database consisting of the epigenetic information of the participants, EEG records and clinical and sociodemographic information of a total of 100 subjects. The value of this database is enormous and will serve both to increase the current know-how and for the development of future projects relevant to both regions (North Portugal and Castile-Leon). — Publication of the results obtained in journals of high impact index and in congresses of national and international relevance.

Objetivos: El objetivo general del proyecto consiste en diseñar una herramienta basada el análisis de epigenética (mediante muestras de sanguíneas) y de la actividad cerebral (mediante el electroencefalograma, EEG), que caracterice la migraña crónica y episódica, simplificando y mejorando su tratamiento. Dicha herramienta será construida a partir del análisis de muestras de sangre y registros de la actividad eléctrica del cerebro. Ambas son técnicas de muestreo rápidas y no invasivas o mínimamente invasivas para el paciente. En la literatura científica, se pueden encontrar varias investigaciones centradas en la genética de pacientes con migraña. Sin embargo, apenas se ha estudiado la epigenética, esto es, los mecanismos de regulación de la expresión génica sin modificación del código genético. Dichos mecanismos de regulación están altamente influenciados por factores ambientales, los cuales también podrían estar relacionados con cambios en la actividad neuronal. Por ello, sería muy novedoso estudiar los cambios que produce la migraña en la actividad eléctrica del cerebro, que se registra mediante el EEG y relacionar dichos cambios con la epigenética. La herramienta desarrollada basará su precisión en la correlación y la información conjunta entre la información epigenética y la actividad eléctrica del cerebro. Esta investigación pretende facilitar la integración laboral y la inclusión social igualitaria. Además, se fomentarán creación de redes y relaciones de cooperación entre entidades transfronterizas para determinar los sustratos epigenéticos y neuronales subyacentes de la migraña. Las entidades participantes se beneficiarán de la posible financiación para mejorar sus equipamientos actuales orientados al trato con los pacientes con migraña, así como al procesamiento y análisis de las muestras. Por otro lado, se beneficiarán con la contratación de recursos humanos y la formación especializada de los mismos en el estudio de esta enfermedad. En relación con el objetivo específico planteado por el programa, se espera que con la creación del partenariado se sienten las bases de una red de excelencia científica en el estudio de la migraña. A este respecto, la divulgación de los resultados derivados del proyecto en congresos nacionales e internacionales, así como la publicación de artículos en revistas internacionales indexadas, mejorará el prestigio y la competitividad de las entidades participantes de manera individual y como partenariado. De manera complementaria, este proyecto también considera la apertura de vías de cooperación adicionales entre las entidades participantes para futuros estudios, como son aquellos relacionados con el tratamiento de esta enfermedad tan expandida en nuestras regiones. Resultados: Los resultados globales del proyecto son los siguientes: - Incremento del número de investigadores en la región de Norte de Portugal y Castilla y León. Se pretende contratar al menos cuatro nuevos investigadores gracias a la financiación proporcionada por esta convocatoria. El nuevo personal se dedicará completamente al proyecto, fomentando la interacción entre las entidades participantes, que se llevará a cabo mediante visitas y estancias en las mismas. - Mejora del equipamiento necesario para el estudio de la migraña en las entidades participantes: equipos informáticos, electroencefalógrafo portátil y material de laboratorio (primers, kits para análisis de metilación, kits PCR, qPCR y RT-PCR, etc). - Creación de una nueva base de datos formada por la información epigenética de los participantes, registros EEG e información clínica y sociodemográfica de un total de 100 sujetos. El valor de dicha base de datos es enorme y servirá tanto para aumentar el actual know-how como para el desarrollo de futuros proyectos relevantes para ambas regiones (Norte de Portugal y Castilla y León). - Publicación de los resultados obtenidos en revistas de alto índice de impacto y en congresos de relevancia nacional e internacional.
